The first settlement in the area was in 1630, when a group of fishermen and farmers settled on Penikese Neck, an isthmus between Cape Cod and the mainland. The town was incorporated in 1798 and rapidly grew due to its central location on Cape Cod and its access to the fishermen's Port of Yarmouth. The town name was changed to Yarmouth in 1819.
