Sister is located in the southern part of Minnesota on the upper Minnesota River watershed. The towns are surrounded by agricultural land. Sister is in the southern tier of the Upper Midwest and has a humid continental climate.
The location of Sister is unusual. It lies in the Driftless Area, a region of interior North America that remained largely unaffected by the last glaciation. There are several lakes nearby, including Mirror Lake, Ellison Lake, and Cedar Lake. The towns are close to Northwoods Minnesota and Whitefish Lake.
The townsite was laid out in 1868. The location was chosen for the railroad, and because of its proximity to water transport on the upper Minnesota River. The townsite was named for two of the founders, Elizabeth Cady Stanton and Susan B. Anthony.
