Depew is a city located in Erie County, New York, in the United States. As of the 2010 census, the population was 35,493. Depew is the most populous city in Erie County and the fifteenth most populous city in New York State. The city is named after DeWitt Clinton, the ninth Governor of New York.

Located at the southeastern corner of New York State and at the southern tip of Lake Erie, Depew is situated approximately sixty miles (100 kilometers) north of Buffalo and eighty miles (130 kilometers) south of Rochester. This city is part of the Greater Buffalo metropolitan area and is located within the Western New York Statistical Area.

The city is surrounded by the towns of Cheektowaga, Lancaster, North Tonawanda and East Aurora.

The city is located in the northeast corner of the state on the bank of the Buffalo River. Depew is home to the Buffalo Military Medical Centre and the Erie County Veterans Memorial.
